Abstract

Objective To detect the changes of interleukin 1 beta (IL-1β) and interleukin 6(IL-6) level in the brain tissue of mice infected with enterovirus type (EV71),and to explore the possible pathogenesis of central lesion caused by EV71 infection. Methods Totally 120 one-day-old ICR mice were divided into infection group and control group.EV71 infected mice model was established by intraperitoneal injection.Fluorescence quantitative RT-PCR technique was used to detect the viral load of EV71 in the brain.Pathological changes on brain tissues were observed through hematoxylin-eosin staining.Enzyme linked immunosorbent assay (ELISA) was used to detect the content of IL-6 and IL-1β in brain homogenate. Results The level of IL-1β was significantly higher in infection group at 3,5 and 7 days compared with that in control group (P<0.01).The level of IL-6 was significantly higher in infection group at 3,5,7 and 10 day compared with that in control group(P<0.01).However,no significant difference was observed in the level of IL-1β and IL-6 in infection group at 1,7,10 and 14 days compared with those in control group (P>0.05). Conclusion The content of IL-1 and IL-6 in brain tissue of mice infected with EV71 was significantly increased,and the immune response mediated by IL-1β and IL-6 may be one of the possible mechanisms of CNS damage.
